Bretazenil, also called Ro16-6028 is an imidazopyrrolobenzodiazepine drug belonging to the benzodiazepine class, classified as a high potency benzodiazepine that binds to α1, α2, α3, α4, α5 and α6 containing benzodiazepine GABA receptors and is a partial agonist. The drug was first synthesized in 1988 and its structure is the closest to flumazenil, but it has slightly different pharmacological properties and is less addictive than other benzodiazepines.
Bretazenil was never marketed as a medicine although it was suggested to have the fewest side effects such as irritability, loss of coordination, liver strain, nausea and aggression.
The dosage, 0.5 mg of bretazenil has about the same psychomotor effects as 10 mg of diazepam. In combination with alcohol, bretazenil creates a sedative-hypnotic effect.
